There are two routes to follow. From Manali to Leh, it takes approximately 12 hours, whereas from Srinagar to Leh, it is approximately 10 hours. The route is: Delhi - Srinagar - Sonamarg - Kargil - Likir Gumpa - Gurudwara Pathar Sahib - Leh. From thereon, you can also explore Nubra Valley and Buddhist Monasteries before returning home. On the other hand, Manali Leh Highway is considered one of the dangerous roads, and the journey is said to be long and tiring, which you would want to avoid at any cost.
